dc.description.abstractThe objective of this project is to develop an automated device used to spot protein samples on a hydrophobic membrane. We have developed two automated methods using a pneumatic tool to bring the pipette to its desired position relative to the hydrophobic membrane. They consist of ways of dispensing the protein complexes on the membrane either by using a high force solenoid or preloading the pipette slowly using the actuator method. The goal of the automated device is to effectively spot the protein complexes such that they are able to be completely separated using 2-D electrophoresis. The automated methods proposed we believe will help prevent the protein binding found with traditional methods. The automated methods will hopefully allow for a simpler and more effective procedure whose end result is the complete separation of proteins without streaking on the membrane surface. It is believed that the high force of spotting the protein onto the surface may allow trapped air in the membrane to prevent the binding of proteins on the surface thus allowing the complete separation of proteins using the 2-D electrophoresis established in U.S. patent 7,326,326. Based on the position and the orientation of the proteins on the membrane, one can detect cancer earlier and determine stages of diseases.
